Lung pathologies are still challenging to diagnose and treat
, despite advancements. Computed tomography (CT) is now the most effective clinical imaging method, yet it has a number of drawbacks.

Spectral photon counting Scanner (SPCCT) is a new imaging modality with advantages over CT and has demonstrated its interest for lung pathologies.

However, new contrast agents must be formulated in order to fully use the capabilities of SPCCT and improve lungs imaging.

In addition to the need for better monitoring of lung diseases, better therapy is also required.

Indeed, the administration of drugs in the lungs faces the problem of crossing different physiological barriers that limit the delivery of therapeutic actives to the target areas in sufficient doses while limiting the adverse effects.

Nanosystems are of diverse interest for pulmonary drug delivery.

They can improve drug stability and solubility, lead to targeting and controlled and sustained release, protect pulmonary clearance mechanisms and improve drug delivery.


The goal of this project is to develop a nanoplatform for simultaneous efficient SPCCT imaging contrast and drug delivery. The combination of contrast agents with therapeutic agents would allow the quantification of the delivered dose of the active agent, the study of its distribution in the lungs and the monitoring of the effectiveness of the treatments.

The major part of the PhD will consist on the formulation of nanoparticles. After formulation, the physicochemical properties will be studied (size, drug encapsulation and release, stability, interactions with biomolecules, etc.). Various analytical technics will be used in collaboration with ICSM (CEA, France). _In vitro_ toxicity of formulations will also be evaluated on cells models of interest.

_In vitro and in vivo_ imaging efficiency will be evaluated in collaboration with cardiovascular radiologist (CREATIS lab, Lyon) on small animals.

The PhD student will be trained for _in vitro_ and _in vivo_ work if necessary.

The PhD will be in collaboration with cardiovascular radiologist (CREATIS laboratory, Lyon) and physico-chemists (CEA, France):

  • ICSM (Institut de Chimie Séparative de Marcoule, UMR 5257 CNRS, CEA, UM, ENSCM). The laboratory have a pioneering expertise in ion-specific effect analysis of nanometric sized ions (nanoion) in solution_ _and in the physical chemistry of colloidal matter and soft nano-assemblies investigated using light, x-ray or neutron scattering.
  • CREATIS (Centre de Recherche en Acquisition et Traitement de l'Image pour la Santé, CNRS UMR 5220, Inserm U1294, UdL, INSA Lyon, UJM). CREATIS addresses imaging and identification of barriers in biomedical imaging (signal and image processing, modelling and numerical simulation) regarding major health issues. The laboratory have a strong expertise in _in vivo_ evaluation of contrast agents with the SPCCT scanner.
