Giant viruses (GVs) encode thousands of protein-coding genes. 70% of such genes encode proteins unseen in any other organisms and thus, are ORFan genes. Such genes likely evolved to hijack cellular functions and outcompete other Acanthamoeba pathogens. Concordantly, their potential use for therapeutics, bioengineering, or methodological advances is tremendous.

During the past two years, we have introduced a battery of tools for the manipulation of the genome of giant viruses.

We now aim to utilize those tools to generate a genome-wide platform of knock-out viruses to assess the function of the approx.

1000 genes encoded by mimivirus.

For this PhD project, the applicant will generate a platform of viruses containing a knockout for each gene of mimivirus and use them to address each gene's overall fitness-conferring phenotype and investigate gene function at different stages in the infectious cycle of mimivirus.

In addition, we will use this platform to identify genes associated with competition between pathogenic organisms by forward genetics. Forward genetic screens will be performed in the presence of competitors like pandoravirus (viral) or Babella spp. (bacteria); or infectious agents like the sputnik virophage, which inhibits mimivirus replication.

Overall, the project presented here aims to provide broad-based functional information on mimivirus genes and facilitate future approaches to utilize viral genes for biotechnological purposes.
